SS-31 (elamipretide) is a synthetic aromatic-cationic tetrapeptide of the Szeto-Schiller family, engineered from an opioid-peptide analog but reworked to eliminate opioid activity while retaining an aromatic-cationic motif for mitochondrial targeting. It carries a net positive charge and alternates cationic and aromatic residues.
It is a focused reference in longevity study, characterized for its association with cardiolipin and its role in models of mitochondrial function.

SS-31 does not bind a protein receptor; instead it is characterized as binding directly and reversibly to cardiolipin in the inner mitochondrial membrane, where it concentrates strongly by electrostatic and hydrophobic interactions independent of membrane potential.
By stabilizing cardiolipin it is reported to preserve electron-transport-chain organization and cristae structure, inhibit the permeability-transition pore, and engage Nrf2/SIRT1/PGC-1α signaling — with little effect on healthy mitochondria.
