NAD+ (nicotinamide adenine dinucleotide) is a coenzyme present in every living cell — a dinucleotide that shuttles electrons through central metabolism in its oxidized and reduced forms. First described in 1906, its tissue levels are documented to decline with age.

It is a foundational reference for cellular research, studied for its central role in energy metabolism and redox-signaling models.

Mechanism

Beyond its redox role, NAD+ serves as a substrate for the sirtuin deacylases (associated with mitochondrial biogenesis and metabolic signaling), for PARP-mediated DNA-repair signaling, and for CD38 — a major regulator of tissue NAD+ that rises with age.

It is synthesized through de novo, Preiss-Handler, and — dominantly — salvage pathways, the latter rate-limited by the enzyme NAMPT.
