FOXO4-DRI is a synthetic, cell-permeable D-retro-inverso peptide — assembled from D-amino acids in reversed sequence for protease resistance — derived from the forkhead domain of the FOXO4 transcription factor and fused to a cell-penetrating domain. It was designed as a senolytic research tool.

It is a specialized reference for longevity laboratory work, studied for its interaction with the p53–FOXO4 pathway in cellular-senescence models.

Mechanism

FOXO4-DRI is characterized as a competitive peptide antagonist that disrupts the FOXO4–p53 interaction in senescent cells, binding the p53 transactivation domain and outcompeting endogenous FOXO4.

Liberated p53 is reported to undergo mitochondrial translocation associated with intrinsic apoptotic signaling — a selectivity attributed to the low FOXO4 expression of non-senescent cells.
