Oxytocin is a cyclic nonapeptide hormone and neuropeptide — the first polypeptide hormone ever sequenced and synthesized, accomplished by Vincent du Vigneaud in 1953. It is produced in hypothalamic neurons and differs from vasopressin by only two amino acids.

It is a foundational reference standard in the cognitive category, studied extensively for neuroendocrine-signaling across a range of models.

Mechanism

Oxytocin acts primarily through the oxytocin receptor, a rhodopsin-type G-protein-coupled receptor whose high-affinity state depends on magnesium and cholesterol. Its dominant pathway proceeds through Gαq/11 and phospholipase C to calcium release and PKC signaling.

Secondary pathways characterized in the literature include MAPK/Rho-kinase, PI3K/Akt/eNOS, and inhibitory Gi/Go signaling; the receptor is also reported to form heterodimers with several related receptors.
