Melanotan-2 is a synthetic cyclic heptapeptide analog of alpha-melanocyte-stimulating hormone, synthesized at the University of Arizona for photoprotection research. A lactam-bridge cyclization and a D-phenylalanine substitution make it markedly more stable and potent than the native hormone and allow it to cross the blood-brain barrier.
It is an established reference in the radiance category, studied for melanocortin-receptor signaling and pigmentation-pathway behavior in laboratory settings.
Melanotan-2 acts as a non-selective agonist at the melanocortin receptors MC1R, MC3R, MC4R, and MC5R — all G-protein-coupled receptors — activating Gs-coupled adenylate cyclase and cAMP/PKA signaling.
Downstream effects are characterized as tissue-specific, including MC1R-driven melanogenesis signaling and central MC3R/MC4R engagement associated with energy-balance pathways.
