Kisspeptin is a family of neuropeptides encoded by the KISS1 gene, originally identified in 1996 as a metastasis-suppressor ('metastin'). Mechanism
Kisspeptin binds KISS1R, a class-A G-protein-coupled receptor, coupling primarily through Gαq/11 and phospholipase C to release intracellular calcium and activate PKC, with downstream ERK, p38 MAPK, and PI3K/Akt signaling.
In GnRH neurons this engagement is characterized as producing sustained depolarization and pulsatile GnRH secretion; continuous exposure recruits β-arrestin and paradoxically suppresses the axis.
