KPV is a naturally occurring tripeptide corresponding to the C-terminal fragment (residues 11–13) of alpha-melanocyte-stimulating hormone. Mechanism
Unlike alpha-MSH, KPV is characterized as not binding melanocortin receptors; instead it enters cells via the PepT1 peptide transporter, which is upregulated in inflamed tissue.
Once internalized it is reported to bind Importin-α3 to block nuclear import of NF-κB and to stabilize IκBα — a dual anti-inflammatory action — alongside inhibition of ERK, JNK, and p38 MAPK signaling.
