The Dossier
Thymosin Alpha-1, or thymalfasin, is a highly conserved, acidic 28-amino-acid immunomodulatory polypeptide originally isolated from calf-thymus extract in 1977. It is the N-terminal fragment of the precursor prothymosin alpha, N-terminally acetylated and intrinsically disordered in solution.
It is among the most extensively characterized immune-signaling peptides in the literature and serves as an established reference across the recovery category.

Mechanism
Thymosin Alpha-1 acts as an agonist of the pattern-recognition receptors TLR9 and TLR2 on dendritic cells, recruiting MyD88 and downstream NF-κB and MAPK signaling that has been associated with dendritic-cell maturation and cytokine expression.
The literature also characterizes a dual regulatory role, including induction of indoleamine 2,3-dioxygenase and tryptophan-catabolism signaling linked to immune tolerance.
