Ipamorelin is a synthetic pentapeptide growth-hormone secretagogue derived from the GHRP series, incorporating non-proteinogenic residues and a C-terminal amide. Developed at Novo Nordisk, it was the first secretagogue characterized for its selectivity of growth-hormone release.

Mechanism
Ipamorelin is a selective agonist of the GHS-R1a (ghrelin) receptor on pituitary somatotroph cells. Receptor engagement activates phospholipase C signaling and intracellular calcium release that drives secretory-vesicle exocytosis — a pathway distinct from, yet synergistic with, GHRH signaling.
Characterization noted its selectivity relative to other secretagogues, without the prolactin or corticotropic activation seen with earlier peptides in the class.
