Ipamorelin is a selective agonist of the GHS-R1a (ghrelin) receptor on pituitary somatotroph cells. Receptor engagement activates phospholipase C signaling and intracellular calcium release that drives secretory-vesicle exocytosis — a pathway distinct from, yet synergistic with, GHRH signaling.
Characterization noted its selectivity relative to other secretagogues, without the prolactin or corticotropic activation seen with earlier peptides in the class.
