CJC-1295 (no DAC), also known as Modified GRF(1-29), is a synthetic 29-amino-acid analog of growth-hormone-releasing hormone. It is a tetrasubstituted derivative of the GRF(1-29) sequence, modified at four positions to resist enzymatic degradation while retaining a short half-life and pulsatile signaling character.
It is a standard reference within the growth category, distinguished from the DAC-conjugated form by its lack of the albumin-binding linker.

Mechanism
CJC-1295 (no DAC) is a selective agonist of the GHRH receptor, a class-B G-protein-coupled receptor on anterior-pituitary somatotroph cells. Engagement activates Gs, adenylyl cyclase, cAMP, and PKA signaling associated with pulsatile growth-hormone secretion.
The four amino-acid substitutions — including the D-Ala² position — are characterized as preventing enzymatic cleavage while preserving high homology with native GHRH.
