Mechanism
GLP3-R is characterized as an agonist at three class-B G-protein-coupled receptors — GIPR, GLP-1R, and GCGR — each coupling principally through Gs, adenylyl cyclase, cAMP, and PKA. Adding the glucagon axis to the two incretin axes is what defines the class, and the balance struck between the three is the property the literature examines.
As with the dual agonists, fatty-acid acylation confers albumin binding and an extended circulating profile relative to the native peptides the sequence is modeled on.
